Using the # tag in your tweet is somewhat like the subject line at the top of an e-mail.  By using the hashtag (#) you are denoting what subject you're tweeting about. 

For example: If I were to post a tweet about how I am grateful it's Friday, I would write my tweet and add #tgif (thank God it's Friday) at the end of my post. 

Using hashtags also helps you, and other Twitter users, search for a specific topic.  By typing in a particular hashtag into the Twitter search bar, you will open up all discussions about your particular hashtag.  To learn more about what certain hashtags mean, and what popular hashtags are in use, click here.
