Get ready to welcome Marmaduke into your home and have a cowabarka good time. The beloved Great Dane of Brad Anderson's famous comic strip comes to life in this hilarious comedy with a grand new adventure in California. When Phil (Lee Pace) takes a marketing manager offer from Bark Organic dog food, he uproots the reluctant Winslow family (wife Debbie (Judy Greer), older daughter Barbara (Caroline Sunshine), son Brian (Finley Jacobsen), younger daughter Sarah (Miliana Haines)), along with Marmaduke (Owen Wilson) and Carlos the cat (George Lopez), and moves them from Kansas to Orange County. New in town, both Phil and Marmaduke try hard to fit in their new respective environments.
Phil tries to impress his boss Don Twombly (William H. Macy), who wants to stock every Petco store in the country with Bark Organic. (It is already number one in California.) Phil's job is to come up with an enticing ad campaign to sway the Petco executives. While Phil is at work, Marmaduke gets to stay in the Laguna Bark Park. It is the high school for dogs, packed with all the typical "cliques" including the jocks, top dogs and outcasts. There he meets and falls in love with the beautiful Jezebel (Fergie), Twomby's Rough Collie. Unfortunately, she already has a controlling and violent Beauceron boyfriend named Bosco (Kiefer Sutherland), who has two Miniature Pinscher lackeys Thunder (Damon Wayans, Jr.) and Lightning (Marlon Wayans). The other dogs he meets there include the outcasts Mazie (Emma Stone), Guiseppe (Christopher Mintz-Plasse) and Raisin (Steve Coogan). Australian Shepherd Mazie's heart is immediately won over by Marmaduke, though he is oblivious to it. Guiseppe is a Chinese Crested dog who is afraid of everything. Raisin is a highly intelligent small-statured Dachshund. Together, Marmaduke and his new friends, Mazie, Guiseppe and Raisin, crash Bosco's "pedigrees only" party and leave after witnessing some of the dogs taunting a cat. Later, Marmaduke asks Mazie for advice on how to win over a girl. Mazie misinterprets and thinks he is referring to her.
Meanwhile, being a teenage dog in a new place is not easy and to earn "top dog" status, Marmaduke enlists Carlos' help. Together they stage a fight to show the locals how tough he is. Then Marmaduke enters in a dog surfing contest that was put together by Phil to woo the Petco executives. All goes well with Marmaduke beating the dog surfing champion Bosco, until they get into a fight and shock everyone, including the Petco executives.
Things keep going downhill from there. Marmaduke breaks Mazie's heart when he uses her advice to take Jezebel on a dream date. While the Winslow family are out on Twombly's boat for a "working vacation," Marmaduke throws a party at the house. It quickly turns sour when Bosco crashes the party and exposes the fight stunt between Marmaduke and Carlos. He is humiliated, loses his new friends and is left with a wrecked house. When the Winslow family return home from a bad "working vacation," they are appalled. Frustrated family and work problems, Phil takes it out on Marmaduke and locks him outside in the rain for the night as punishment.
Devastated, Marmaduke decides to run away. The youngster meets a wise elderly "ex-top dog" English Mastiff, who goes by the name of Buster or Chupadogra (Sam Elliott), because it is rumored he killed his owner. Offering some sagely advice, will Chupadogra be able to convince Marmaduke that things are not as bad as they seem? And will Phil be able to find a way to seal the deal with Petco before he gets fired and make amends to his neglected family? Both dog and owner have a lot to figure out if they are going to make it in The O.C.
