Killers is an action comedy starring Ashton Kutcher as the contract killer Spencer Aimes and Katherine Heigl as Jennifer "Jen" Kornfeldt, the woman he meets while he is on a job in Nice. She is vacationing there with parents (Tom Selleck and Catherine O'Hara) after a bad breakup. Although Spencer's life involves guns, bombs, exotic sports cars and beautiful women, he feels unsatisfied with his life. He falls in love with Jen and gives up the business for her. Spencer's boss Holbrook (Martin Mull) disapproves and warns him that he cannot just walk away from the "life."
Spencer and Jen get married and three years later they have settled into a "normal" life together in the suburbs. Jen surprises Spencer with tickets to Nice to celebrate his birthday and their relationship. However, Spencer is not enthusiastic because of his contract killer connections to Nice. Unaware, Jen misinterprets his reaction and her friends help fuel her insecurities about their relationship. (Back in Nice, Spencer had told Jen about his line of work, but she had been drunk and passed out.)
Meanwhile, Spencer receives a postcard from his former boss requesting him to return back to the contract killing business. He calls his former boss and reiterates his refusal to go back to his old line of work. Later that evening, at a surprise birthday party for Spencer, he is not his usual self and seems very distracted. Jen grows more concerned when Spencer rejects her sexual advances the next morning and rushes her off on her business trip.
When Jen returns, she find Spencer and his friend and co-worker Henry (Rob Riggle) scuffling in their living room and the house in shambles. Although shocked, she manages to grab Spencer's gun after some prodding and ends up shooting Henry. Subdued, the truth comes out. Henry confesses there is a $20 million bounty out for Spencer. Confused and unaware of Spencer's past, Jen freaks out and demands an explanation. Their argument is cut short when a sniper tries to shoot Spencer and there are more hitmen after them. Jen and Spencer no longer know who to trust when various familiar faces of people they thought they knew come after them. To complicate things more, Jen is pregnant!
While dodging bullets, Jen and Spencer will have to test their relationship to the max and work together to find out who contracted the kill before they both end up dead.
